Dr. Lorraine Kalia<\/span><\/p><\/div>\n

At the University of Toronto, Dr.\u00a0Lorraine Kalia, a neurologist and neuroscientist, is investigating the role that a protein called alpha-synuclein plays in a familial type of Parkinson\u2019s disease caused by a mutation in the LRRK2 gene. Although large clumps of alpha-synuclein have been incriminated in the death of dopamine-producing brain cells, Kalia is investigating smaller deposits of the protein to see if their misbehaviour contributes to the death of brain cells and patient symptoms.<\/p>\n

Researchers have identified a protein called alpha-synuclein as a major culprit in Parkinson\u2019s disease because it accumulates in large clumps, known as Lewy bodies that are associated with the death of dopamine-producing brain cells.<\/p>\n

Autopsies on the brains of some people with a genetic form of Parkinson\u2019s have revealed they don\u2019t have Lewy bodies.\u00a0 So with this two-year, $90,000 Pedaling for Parkinson\u2019s New Investigator Award from Parkinson Canada, Kalia, is trying to find out why their dopamine-producing brain cells died.<\/p>\n

Kalia has collected autopsy cases of people with genetic Parkinson\u2019s disease, caused a mutation in the LRRK2 gene. She believes people with that form of Parkinson\u2019s may have smaller clumps of alpha-synuclein, called oligomers, in their brain cells.<\/p>\n

\u201cThese smaller aggregates might be the larger culprit for what kind of alpha-synuclein causes brain cells to die,\u201d she says.<\/p>\n

Kalia is comparing the brains of people who did have Lewy bodies\u2014the large aggregates of alpha-synuclein\u2014to the brains of those who did not have those large clumps. She\u2019s looking for the smaller oligomers.<\/p>\n

\u201cThe protein is still building up in brain cells, but not in the same way,\u201d she says. \u201cIt\u2019s still there; it\u2019s still misbehaving, but possibly in these smaller clumps.\u201d<\/p>\n

Once Kalia and her team have determined how large and how many oligomers there are in the brains of people with this genetic form of Parkinson\u2019s, she\u2019ll determine if the way the oligomers are distributed in the brain correspond to different symptoms.<\/p>\n

Kalia\u2019s previous research suggests people with Lewy bodies were more likely to have dementia or other non-motor symptoms than those without Lewy bodies.<\/p>\n

She hopes this research will shed light on whether the location of the clumps of misbehaving proteins, or the type and size of clumps\u2014Lewy bodies or oligomers\u2014correspond to non-motor symptoms.<\/p>\n

Ultimately, Kalia hopes her research will open a new avenue for drug testing or other forms of therapies to treat whichever form of alpha-synuclein is causing the problems.<\/p>\n

Although 75 percent of her time is spent on research, Kalia spends the other 25 percent caring for people who have Parkinson\u2019s.<\/p>\n

\u201cMy exposure to people who have Parkinson\u2019s disease is really what inspired me to pursue a career in movement disorders,\u201d she says. \u201cThe fact that we have a lot of clinical unmet needs in Parkinson\u2019s disease \u2026 is the driving force of the lab.\u201d<\/p>\n
